Project objective
Of the 50 or more structurally distinct phospholipids present in platelets only a few have been quantitatively analysed or shown to participate in cell signalling or other platelet functions. Many lipids can now be analysed by GC-MS and LC-MS and one and 2D-NMR lipid (and cytosolic) profiles have been analysed for all classes of phospholipids. These two complementary approaches will be utilised to elucidate the role of specific lipids and lipid moieties in cell signalling and in the morphological changes induced by agonists and antagonists like collagen, thrombin, PAF, NO etc. Comparison of the profiles from resting and stimulated platelets, when carried out in a time-and-dose-dependent manner, should correlate with shape change, aggregation, exocytosis-degranulation, PAF and eicosanoid formation by different phospholipases A2, C and D. Comparison of the platelet lipid profiles from healthy and diseased volunteers will be used to correlate changes in specific with the severity of platelet associated diseases such as thrombosis, hyperlipidemias, etc.
