Characterization of control elements in the protein 's gene promoter

Project objective

Thromboembolic disease is still one of the commonest causes of morbility-mortality of our society. A significant number of patients with trombosis have a congenital deficiency in one of the proteins involved in the regulation of blood coagulation, such as Protein S (PS). There is a variable biochemical expression of the PS gene in healthy controls and PS deficient patients, which appears to depend on age, sex and tissue. Moreover, during pregnancy or administration of oral contraceptives, the PS levels are dramatically reduced, indicating a possible link between hormones and PS gene expression. In spite of the recent characterization of the PS gene, the 5' region, where the regulatory elements are localized, is still poorly studied. To try to explain the above observations, the aims of this project are the identification and characterization of the functional elements of the PS gene promoter, as well as the study of the control elements responsible for tissue specific expression and the hormonal response. We will also study the transcriptional start site to know whether there is more than one and if found, if are they tissue specific.
