Project objective
My aim is to contribute to the knowledge on structural properties of transfer ribonucleic acids (tRNAs) in general, and to monitor fine conformational differences between wild-type and mutated functionally invalid molecules in particular. Key molecules to be investigated will be (i) mutated yeast tRNA transcripts already explored for their aminoacylation properties, to allow establishment of local conformations, and understanding of the contribution of structure to the expression of identity, (ii) human mitochondrial tRNAs whose point mutations are correlated with severe pathologies. Establishment of the structural characteristics of wild-type mitochondrial tRNAs (a quite unexplored field) and comparison with those of their mutated counterparts will shed light on the way the mutations affect the biological activity. The approaches will include (i) chemical and enzymatic in vitro structu probing of RNA using well established and newly developed probes, (ii) computer modelling, (iii) optical melting investigations, (iv) temperature gradient gel electrophoresis.
