Project objective

In all eukaryotes the centromeres are essential for separation of chromosomes into dividing cells. The centromere DNA forms together with several proteins a large complex called the kinetochore. The role of the kinetochore is to mediate contacts with the microtubules and motor proteins that pull the chromosomes to opposite poles of the mitotic and meiotic spindles. The centromere DNA sequence has been defined in fission yeast but the protein components are hitherto unknown. The aim of this project is to clone and characterise the genes which encode trans-acting components of the fission yeast kinetochore. To this end six temperature sensitive lethal mutations called cspl -6 (centromeric suppressors of position effects) and six nonlethal mutations (csp7-12) have been isolated. These are good candidates for kinetochore mutations because they affect the in vivo accessibility of the centromere DNA and cause chromosome loss.The corresponding genes will be cloned and sequenced. The immunolocalisation of the gene-products will be investigated.
