Project objective

Plasma membrane vesicles will be isolated and used to study the mechanism(s) that controls monovalent anion uptake in the green alga Monoraphidium braunii. These anions (NO3-, NO2- and Cl-) are essential for the development of all plant cells. The activation of this uptake system depends on the presence of blue/UV radiation in the actinic light. Using plasma membrane vesicles, we will attempt to localize, identify and, if possible, purify the photoreceptor(s) involved. Recent experiments suggest that in vivo a cyclic GMP-dependent phosphorylation occurs during the activation of the uptake mechanism. The role of protein phosphorylation in the sensory transduction of the blue/UV light-dependent activation of these transport processes will also be studied. Conversely, a photosynthetic rate approaching saturation is required for the full activation of the entrance of monovalent anions in the cells. The requirements into energy and/or reducing power for the transport of these anions will also be investigated using plasma membrane vesicles.
