Hepatitis g. viruses - epidemiological and clinical study and molecular and serological analysis

Project objective

Viral hepatitis is known to be caused by a number of viruses, each with its own distinct mode of transmission and replication. On April of this year, a new virus has presented in a congrcss in Washington the GBV" (RNA virus, Flavivirus-like) with 3 known subtypes (A, B and C). HGV is another Flavivirus discovered recently which is able to develop hepatitis. The aim of the study is to investigate the medical, epidemiological and pathological behaviour of these new viruses using sera collected in the host laboratory during several years. WORK PROGRAM: 1. To discriminate by ELISA using synthetic peptides immune reactive sera against "GBV" in several risk groups. Also studies concerning the GBV transmission will be performed. 2. RT-PCR method will be used to detect HGV or HGV genome presence in sera, liver and lymphocytes in patients diagnosed of non A-E hepatitis. Positive and negative strands will be investigated. 3. The effect of interferon treatment on HGV or GBV replication will also be studied retrospectively by PCR in patients with HBV or HCV coinfection. 4. Finally, the role of these viruses in hepatocarcinoma and cirrhosis will be investigated. This is an important opportunity for the proposer and the host laboratory to contribute at the understanding of a problem of pivotal relevance for public health in the world. With this project, we will contribute not only to further knowledge about hepatitis viruses but also our hospital will be one of the first hospitals in Spain and Europe in which GBV and HGV tests will be available with a lot of consequences in the sanitary system.
