Project objective

The development of T and B lymphocytes characterized by a precise sequence of maturatlon steps, some of which defined by the expression of specific cell surface markers. Aim of this research project is to develop a system for the analysis of the molecular and cellular aspects during the early phases of lymphoid development : the first effort will be devoted to the establishment of reproducible system for generating T and/or B lymphocytes from murine embryonic stem cells (ES) in vitro. A combination of varius cytokines and several stromal cell lines known to have a potent lymphohematopoietic promoting activity will be used on these undifferentiated, pluripotent cells, both to elucidate the relative role of these cytokines in lymphoid development and to better understand T cell differentiation Secondly, this system will be used to identify new genes involved in the lymphoid differentiation pathways, using a strategy based on the gene-trap approach.
