Project objective

During the years a number of specific chromosome translocations have been identified in several classes of soft tissue sarcoma. In Dr. Coopers laboratory the translocation t(X;18)(pll.2;qll.2) found in synovial sarcomas has recently been identified. They showed that during this translocation fusion genes are created between the SYT gene on chromosome 18 to either SSX1 or SSX2 on the X chromosome. These genes were also cloned and were identified as novel genes. In the project that I will work in, the normal functions of the SYT, SSX1 and SSX2 proteins will be studied. The mechanism of the SYT-SSX1 and SYT-SSX2 fusion proteins in cellular transformation will also be studied. A third aim of this project is to investigate the possibility to use antibodies against SSX1 and SSX2 proteins in tumour diagnosis.
