Project objective
The aim of this research project is to isolate pharmacologically GABA-mediated postsynaptic currents in the nucleus accumbens evoked by stimulation of this structure and to characterise the inhibition of this synaptic input by dopamine. The dopamine receptor involved will be identified. We will study the cellular mechanism involved in the action of dopamine on this current and the presynaptic action of dopamine. I will use and learn whole-cell patch-clamp recordings in brain slices in the laboratory of Dr. M.G. Lacey where this technique is routinely used. This technique is complementary to the ones I already know and is useful to investigate other aspects of dopaminergic transmission in the brain. The experiments suggested in the research project will help us to answer the following questions: + Is dopamine depression of inhibitory postsynaptic potentials a direct action on GABA neuron tenninals? * Is the dopamine receptor involved of the D1 type? * Does the mechanism involve stimulation of adenylate cyclase?
