Project objective
Research objectives and content GABAergic interneurones show a large variability in the repertoire of receptors present on their cell surface. Furthermore networks of GABAergic interneurons generate synchronized '40 Hz oscillation' modulating the populations of the principal excitatory cells, a phenomenon underlying the mechanism by which the cortex can associate different features of an object a unified perception. The aim of this research project is the generation of transgenic mice in which one of the GABAA receptors or metabotropic glutamate receptors is selectively knocked out in a specific neuronal population, the GABAergic interneurones, to evaluate if the synchronized 40 Hz oscillation" and the cognitive mechanisms are consequently altered. This result will be achieved using innovative techniques that implies the generation of multiple transgenic mice lines, that can be further used for generating different genetically modified mice lines. Training content (objective, benefit and expected impact) This training period will allow me to learn advanced molecular biology techniques, such as generation of transgenic and knock-out mice, and will complete my scientific experience on the cognitive mechanisms field adding a new focus on the role of the GABAergic neurons. "
