Project objective

Research objectives and content Axonal pathfinding in the nervous system is mediated in part by cell-to-cell signaling events involving members of the Eph receptor tyrosine kinase family and their membrane-bound ligands. The research objective is to establish the role of the ligand EphrinB 1 in the pathfinding of neurons within the mammalian nervous system. The first part of the research plan is to knock-out the Ephrin-B 1 gene to establish the significance of this ligand in directing axon guidance as well as the involvement of this geneproduct in border formation and migration of nerve cells. In a second project, we will produce a knock-in construct in which exonic sequences of Ephrin-B 1 will be replaced by an Ephrin-B 1 cDNA lacking the cytoplasmic domain (EphrinB 1-DeltaC). Analysis of such mice will supply genetic evidence for the importance of Eph-receptor induced ligand-signaling by use of the cytoplasmic domain of the ligand. Training content (objective, benefit and expected impact) The objective is to receive experience in the generation of genetic models to investigate complicated molecular interactions in the central nervous system. Several benefits are at hand, such as the establishment of fruitful interactions and the first-class training in an expanding and innovative area of neuroscience research. The training is expected to significantly increase the applicants knowledge in the field of neuroscience, and in particular of the organisation and requirements of cell-to-cell signaling in the central nervous system.
