Mechanism of action of estradiol in the neuronal control of the preovulatory lh surge in the ewe

Project objective

Research objectives and content The luteinizing hormone-releasing hormone (LHRH) neurones of the brain control reproductive cycles in the female. Although estrogen, secreted by the ovaries, exerts an important influence on LHRH neurones it is not known how this occurs in any species. The objectives of this project are to: 1) Define the neurones expressing estrogen receptors which are likely to project to the LHRH neurones of the ewe 2) Determine the neurotransmitters being used by these neurones, and 3) Establish the effects of estrogen on neurotransmitter synthesis in these cells. The project will involve the use of retrograde tracing techniques, double- and triple- labelling immunocytochemistry to achieve 1) and 2), and in situ hybridization for the analysis of specific neurotransmitter-related messenger RNAs in the brain of the ewe Training content (objective, benefit and expected impact) The objective of my proposed post-doctoral study at The Babraham Institute will be to provide myself with a solid grounding in 'state-of-the-art' neuroanatomical and cellular messenger RNA techniques. This will complement my present skills regarding in vivo neurobiological assessment and is critical if I am to obtain the global training now required within the field of neurobiology. A successful post-doctoral period in Dr Herbison's laboratory should greatly enhance my chances of acquiring a University position in France.
