Project objective

The MAP kinase pathway regulates fundamental processes such as mitogenesis, transformation, apoptosis, inflammation, and neuronal functions. It is unclear how one pathway can fulfill these diverse tasks. We want to specify the role that B-Raf kinase plays in determining its set of substrates and cell reponses of the MAPK pathways. The host laboratory has considerable experience working on the Raf->MEK-> MAPK/ ERK pathway, the partner proteins associated with the kinases and analysis of cellular Reponses. The facility to merge proteomics with genetics in order to address receptor signaling cascades and cell responses gives the applicant a rare opportunity and will place him in a excellent position to successfully continue with the project in the future. High profile research on MAPK signaling provides a significant biological question worthy of this technology. The institute also provides invaluable experience in many other aspects of cell signaling. The applicant will provide expertise in cloning, expression and purification of fusion tagged proteins, experience in the analysis of molecular interactions between signal proteins. Having completed a short-term research associate in the recently established proteomics field, illustrates that the applicant provides the skills pertinent to the project. Thus, signaling complexes, via proteomics, and functionality of constituents, via utilizing protein variants known to affect the pathway signaling, can be addressed.
