Project objective
A newly developed method is available now for routine use to fractionate alive complex bacterial communities from a diverse range of habitats to the species level (BAC-PACE). It is therefore the first and only method available at the moment, which allows the live isolation of single bacterioplankton populations, which can then be used for metabolic and physiological measurements and for biotechnological and pharmaceutical screening. The diversity and kinetics of bacterial ectoenzymes can be assessed by capillary electrophoresis (CERES) with an unsurpassed sensitivity. Thus isolation of bacteria and enzymes and their characterization can be directly done on environmental samples without the need of cultivating them first. Ultimately, sequencing will provide information on the phylogeny of the isolated bacterial species on which the various measurements were performed. All these mentioned techniques would be introduced both theoretically and by extensive hands-on instructions. Good background in microbiology and basic knowledge in chemistry is required.
