Project objective

The training area consists of advanced methods for characterizing microbial strains which will provide a basis for understanding the behavior of hyper producing strains used in industry for production of commercial products like industrial enzymes. Comparison of such strains to laboratory strains has been hampered by the lack of genetic markers, a sexual life cycle, and developed methods for para-sexual crossing, so even though extremely high yielding strains are used in industry, the basis for this is largely unknown and can therefore not be used when production strains are developed for new products. Recently new technologies have been developed which provide new information on how organisms behave. One such technology is C-13 labeling in combination with total hydrolysis of biomass and separation of monomers by GC-MS. By looking at the molecular weight of molecules and parts of molecules, the C-13 label can be followed, and this information is used to verify or discharge suggested metabolic pathways operating in the cell. Another new technology is based on DNA micro arrays, where the expression of in principle all genes, can be monitored in parallel. This technology is particularly powerful to compare two strains grown under identical conditions or to compare two sets of conditions used for growing one strain. By applying these two techniques on hyper producing industrial strains new and very exciting information on why these strains behave as they do will definitely emerge.
