Final Activity Report Summary - INFO-FOCT (Information in Functional Optical Coherence Tomography and Microscopy)

The objectives of the 'Information in functional Optical Coherence Tomography' (Info-fOCT) project were: 1. a thorough theoretical and experimental exploration of light interactions within tissue using coherent light imaging approaches; and 2. the establishment of new measuring platforms based on optical coherence tomography and spectroscopy for imaging function and structure in biomedical settings. The main achievements that resulted from this project were: 1. the development of advanced simulation tools to efficiently study light propagation through tissue; 2. the evaluation of the information limits of optical coherent imaging-based therapies through tissue; and 3. the development of unique optical technologies for biomedical imaging at various resolution scales ranging from a few micrometres to a few nanometres.

Project objective

The full potential of coherent-gated functional imaging capabilities in scattering media has not been yet realized. Particularly, optical coherence tomography and microscopy (OCT) present a promising investigational tool in cellular biology, especially when combined with principles of Information Theory for analysing the measured functional OCT (fOCT) signal. This research aims to explore the dynamic non-linear response of tissues and cells excited by an optical pump signal by means of optical coherence imaging and spectroscopy. Sophisticated path-length and transverse-direction-resolved studies of measures such as water absorption and polarization states of reflected light will be conducted.A theoretical analysis will be directed towards:(i) deriving the Volterra kernels associated with the non-linear response of the biological medium as well as performing importance sampling Monte-Carlo light-tissue simulations in order to understand the relationship between the medium properties and the measured fOCT signal;(ii) the use of Shannon and Fisher information quantities to determine the information amount conveyed by the fOCT signal.The research results not only will set the theory of the inverse problem in fOCT on a firmer foundation, but also will significantly affect the safety and optimisation of the optical feedback control of therapeutic lasers, for instance in laser ablation procedures. This interdisciplinary proposal brings together Prof. Brett Bouma's group at Harvard Medical School and Prof. Theo Lasser 's group at the EPFLausanne, to combine their extensive expertise on OCT and microscopy with the objectives of adding scientific and complimentary competencies to the career of the applicant, Alberto Bilenca, extending his international research experience and providing him with new knowledge in the area of functional optical coherence imaging, which will be gained by the EPFL group, as well as will enable Alberto to establish his own academic career.
