Project objective

The MAPKINASE project aims to elucidate aspects of the mode of action of the important plant growth substance ethylene by integrating genetic, biochemical and proteomic approaches, hence capitalising on the post-genomic advantages of Arabidopsis as a model system. The scientific objectives are two-fold. Firstly, to define the ethylene-modulated phosphorylome and secondly to model phosphorylation flux in ethylene-mediated MAPkinase cascades. The work will use tissue from wild type treated with or without et hylene and will also study the ctr1-1 mutant where the lesion is in a MAP3kinase gene, (phenotype is constitutively ethylene-treated") and where the relevant cascades are switched on. The work will deploy advanced techniques in proteomics including two-dimensional electrophoresis and differential staining with fluorescent dyes to identify differences with treatment, together with a range of mass-spectroscopies for identification, combined with advances bioinformatics. The outcomes will result in a much d eeper understanding of the processes of signal transduction. The proposal responds to the need to bring to the research project, the top class scientific expertise of the proposed Fellow whilst also providing opportunites for continuing professional develo pment including access to an independent professional mentor and training courses in ethical and financial management. The broader objectives include training in a number of relevant scientific and complementary skills including developments in protein p urification separation and detection, robot-technology, mass spectrometries and statistical analyses of proteome data-sets. The research, in support of the Lisbon and Gothenburg objectives, will not only contribute to the developing knowledge-based economy but will aid the development of genuinely effective and working relationshiops with Russia. The project fully supports the mobility agenda of the ERA to encourage the spread of knowledge within Europe."
