Project objective

Single nucleotide polymorphisms have become the 'industry standard' marker type in mammalian and human genetics, and will be important for a biotechnology-oriented approach to genotype improvement and redesign. The project aims to develop a generic platform for genic SNP discovery in the major temperate cereal crop species, bread wheat. Polyploidy compromises the standard approach for sequence variation detection in specific amplicons, and to address this, a strategy using SSCP separation of complex amplicons to develop homoeologue-specific PCR assays is proposed. The proposal will exploit the rice genome sequence and the existence of synteny among the grass genomes to drive primer design, and this is complemented with a SNP discovery platform based on heteroduplex identification using dHPLC technology.
