Project objective
The proposed research concerns the synthesis of orthogonal  differentially binding  peptide interaction pairs for the in vitro and in vivo labelling of proteins. The complex formation of these peptide pairs is based on multiple coordinative interactions between a polyhistidine peptide on the one side and a complementary peptide bearing multiple Nickel-nitrilotriacetic acids (Ni-NTAs) on the other. Multivalent Ni-NTA peptides will be prepared by solid phase peptide synthesis using synthesized NTA amino ac ids. Complementary histidine peptides, selected by screening a library of polyhistidine peptides with the NTA peptides, will be genetically engineered as fusions to proteins.The most promising orthogonal peptide interaction pairs will be applied for the simultaneous, site-specific and differential labelling (in vitro and in vivo) of various histidine-tagged proteins with different fluorophores coupled to the NTA peptides. It is envisioned that this unique orthogonal labelling technique will offer a new means for monitoring protein interactions and conformational changes in real-time.
