Накратко на български

Специални полимери и протеини се използват за създаване на частици, подобни на вируси, които могат да транспортират лекарства в организма. Те помагат за по-точното доставяне на терапия и подобряват качеството на изображенията при магнитен резонанс.

Self-Assembled Virus-Like Particles from Polyferrocenylsilane-Based Polymers and Viral Capsid Proteins: Fabrication, Surface Engineering and Applications

Work progress and achievements during the period February 01, 2017-January 31, 2018 Goals and key milestones: a) Construction of VLP–based drug delivery systems. b) Investigating the MRI enhancement and controlled release behavior of VLP–based DDSs. c) Preparing research articles and applying patents based on the results discovered. d) Conferences attendance. e) Outreach activities. f) Knowledge transfer. The research has progressed based on the schedule (see below). a) VLP-based drug delivery carriers were constructed via the self-assembly of polyferrocenylsilane-based copolymers (PFSs) containing hydrophilic functional groups and hydrophilic biomacromolecules, which include polysaccharides, amino acid, and proteins. Besides, the mechanism of the assembly process was elucidated. On this basis, several hydrophobic drugs were encapsulated into the carriers. b) The MRI enhancement capability and controlled release hehavior of VLP-based DDS were investigated systematically. c) The results as above have been drafted three papers, which have been published repectively in Nature Communications, Acta Biomaterialia, and Chinese Journal of Polymer Science during the project period. The papaers are listed as follows. (i) Hao Wang, Tingting Dai, Shengli Li, Shuyan Zhou, Xiaojing Yuan, Jiayi You, Chenglong Wang, Vincent Mukwaya, Guangdong Zhou, Guojun Liu, Xiaohui Wei, Hongjing Dou*, Scalable and cleavable polysaccharide nanocarriers for the delivery of chemotherapy drugs, Acta Biomaterialia, 2018, 72, 206-216. (ii) Hao Wang, Ting-Ting Dai, Bo-Lun Lu, Sheng-Li Li, Qing Lu, Vincent Mukwaya, and Hong-Jing Dou*, Hybrid Dextran-gadolinium Nano-suitcases as High-relaxivity MRI Contrast Agents, Chinese J. Polym. (i) Dr. Dou participated in teaching a postgraduate course on "Supramolecular Materials and Biomedical Applications" in November 2017 at SJTU, the course was taught in English and covered both native and oversea students. (ii) Dr. Dou joined the “2017 Shanghai Conference on Scientific Research and Innovation in Europe” on Nov. 30, 2017, and communicated with some EU staffs and scientists for further project application and research collaboration. (iii) The paper published in Nature Communications was reported by several Chinese medias, the links are listed as follows: http://baijiahao.baidu.com/s?id=1578243574266206678&wfr=spider&for=pc https://www.xianjichina.com/news/details_48908.html http://www.cailiaoniu.com/101233.html http://www.sohu.com/a/191631261_472924 f) Knowledge transfer: During the period of the project, knowledge of PFS copolymer synthesis and protocell preparation has been transferred to Dr. Dou from the Manners group and the Mann group. New collaboration between the three-parties, i.e., Manners group, the Mann group, and the fellow’s group at Shanghai Jiao Tong University has been established based on the fellow’s work. The fellow co-hosted Prof. Ian Manners’s visit to Shanghai Jiao Tong University and organized a mini-symposium for Prof. Ian Manners on April 11, 2018, three students from Hongjing Dou’s group presented in the mini-symposium regarding collaborative projects.

Цел на проекта

The capsid proteins of viruses have been shown to organize around a variety of non-biological polyanions, in a similar way that proteins assemble around RNA genomes, to form virus-like particles (VLPs). Metal-containing polymers such as polyferrocenylsilane possess additional interesting physical and chemical properties and may yield VLPs possessing very different morphologies and more diverse functionalities on co-assembly with capsid proteins. In a similar manner to almost all nanoscale objects, VLPs generally exist in the solid state but not in the liquid phase, because the scales of these systems are typically larger than the range of attractive interactions between such nanostructures. This situation comes with limitations regarding both storage and product formulation. Due to the recent development of liquid proteins via the surface engineering techniques, interest in the phase behavior of bionanomaterials has grown rapidly. The proposed research focuses on the fabrication and surface engineering of responsive VLPs through the self-assembly of polyferrocenylsilane-based copolymers and viral capsid proteins. This project is highly interdisciplinary and the project objectives will be accomplished by the proposed award of a Marie Curie Fellowship to a highly talented young scientist from China, Dr. Hongjing Dou. She has considerable expertise in the area of bionanomaterials and in biomedical science. The proposal involves her working at the School of Chemistry at the University of Bristol in the UK together with Prof. Ian Manners, who has expertise in the field of synthetic metallopolymers such as polyferrocenylsilanes and also self-assembly, and cosupervisor Prof. Stephen Mann, an expert in bio-inspired chemically-derived routes to complex materials and a pioneer of solvent-free liquid proteins and viruses, to achieve the ambitious project goals.
