Genetic interactions describe the phenotypic consequences of variations in a specific gene, whether genetically or environmentally induced, associated with other gene. Discovering synthetic genetic interactions is crucial for unraveling the complexity of metabolic pathways and cellular functions, offering deeper insights into how cells operate under stress. Traditional genetic studies in C. reinhardtii have often focused on individual gene mutations, without fully exploiting its sexual reproduction capabilities to generate double mutants. This project aims to bring a new perspective to the study of gene functions in C. reinhardtii by identifying synthetic genetic interactions among nutrient metabolism genes. Through an innovative interdisciplinary approach involving the use of available single mutant strains from Chlamydomonas Library Project (CLiP) collection, established genetic tools, and the generation of haploid double mutants via crossing, this project seeks to uncover complex interactions between metabolic pathways. By serving as a foundation for future large-scale synthetic interaction studies, this project goes beyond current methodologies in studying genetic functions in C. reinhardtii, impacting the research field of metabolic regulation in photosynthetic organisms. The RAFAEL project will contribute significantly to the fellows professional development, equipping him with expertise in algal genetics and expanding his interdisciplinary research scope.
