The objectives of this proposed project is the elucidation of the metabolism and potential novel second messenger roles of the glycerophopshoinositol and lysophosphatidylinositol, the principle products that originate from phgosphoinositide-specific phospholipase a2 (pla2) activation. This enzyme has recently been characterised in thyroid cells and leads to the formation of these biologically active metabolites (Iacovelli et al, 1993; Corda et al, 1994; Falasca and Corda, 1994; Falasca et al, 1995). This should thus provide a further understanding of the proposed mechanisms of cross-talk between adenylylcyclase, PLA2 and the tyrosine kinases, particularly in the light of recent reports of both potentiating (Sevetson et al, 1993) and inhibitory (Wu et al, 1993) intercations between adenylylcyclase and the tyrosine kinases (Burgering et al, 1993; Cook and McCormick, 1993) that could be under feedback control via MAPK stimulation of PLA2 (Graves et al, 1993; Durstin et al, 1994), and hence via the production of glycerophosphoinositol 4-phosphate (Valitutti et al, 1991; Iacovelli et al, 1993).
