Цел на проекта
The secretory pathway of eukaryotic cells comprises several specialized membrane-bound compartments. Transfer of proteins between these compartments is mediated by transport vesicles. A correct targeting of these vesicles is accomplished by interactions between integral membrane proteins on the vesicles and target organelles, termed v-SNAREs and t-SNAREs, respectively. SNAREs required for anterograde transport from the endoplasmic reticulum (ER) towards the plasma membrane are already known. The discovery of a novel t-SNARE on the ER of yeast allows us to investigate for the first time the targeting of retrograde moving vesicles in the secretory pathway. Since both anterograde and retrograde v-SNAREs must be present on the same vesicles, either as targeting molecules or recycling passengers, it is possible that some of the vSNAREs involved in forward transport also have a role in retrograde traffic. The genetic and biochemical studies described in this proposal offer a unique opportunity to determine whether any of the known v-SNAREs act in this way or whether there is a separate set of retrograde vSNAREs to be discovered. Our investigations should contribute to a better understanding of how eukaryotic cells maintain their compartmental organization.
