Цел на проекта
The processes of cellular proliferation are regulated through the activation of specific signal trasduction pathways resulting in alterations of the gene expression pattern. Thus, the ultimate targets of these pathways are represented by nuclear factors which bind to specific enhancer sequences and integrate information from promoter sequences and different extra- and intracellular signals at the level of transcriptional control. Among these factors, several proto-oncogenes have been described. Altered regulation of proto-oncogenes may lead to unrestrained cellular growth, wich is a remarkable feature of the transformed phenotype. Several data indicate that CREM proteins, nuclear factlors belonging to the leucine zipper proteins family, are able to regulate the expression of some oncogenes, i. e. jun and fos, both at transcription and post-translational levels. In this perspective, the overall aim of this project is to investigate the possibility that CREM proteins might modulate cell proliferation by affecting the activity of immediate-early response proto-oncogenes. In order to adress this question, several experiments will be performed both in cultured cells, by the use of transformation assays, and in the whole organism, by the establishment of transegenic mouse lines, to assess whether CREM proteins might affect in vivo tumor induction.
