Цел на проекта

Molecular cytogenetics involves studying the physical and genomic organization of repetitive and single copy DNA sequences. The first part of this research proposal extends the work started at the John Innes Centre by the applicant isolating DNA sequences from Avena species, and study their organization on diploid, tetraploid, and the cultivated hexaploid oat species, as well as in synthetic octoploids. The genus Arrhenatherum, the most closely related genus to Avena, will also be included in the study. This part will complete the study of organization and architecture of repeated sequences present in the four different genomes (A, B, C, and D) in oats, and the types of sequence evolution that have occurred during speciation of the genus. The second part involves the molecular cytogenetic analysis of Olea europeae, a major crop in the country of origin of the applicant. Little is known on the origin and the evolutionary stages of the cultivated olive; furthermore, there is little cytogenetic information for the Olea genus. We propose to investigate the genomic structure of 0. europeae and compare it with other Olea species. This analysis will help to clarify the evolution of the cultivated olive in relation to some suggested progenitors, such as 0. chrysophyllum, and 0. excelsa. We will also try to differentiate the Greek olive varieties by molecular DNA techniques. The aim in part three is to physically map the inserted gene(s) in transgenic rice lines. The project is based on experience acquired by the applicant during his 2-year training period at the Karyobiology Group at the John Innes Centre.
