Цел на проекта
Each team will train a young post-doctoral fellow from an EC member state. major histocompatibility complex (MHC) with peptides derived from protein Importantly, the post docs will integrate into the training programs of the antigens. Therefore, the objective of the project is to unravel the home institutes, and work in selected laboratories of network partners for additional expertise. Additional important measures are annual meetings mechanism leading to peptide loading of MHC class I and class II (2-3 days) attended by all young researchers in the network, and mutual molecules. Part of the project became possible through the recent pioneering discovery by J. Trowsdale and other laboratories that the MHC short-term visits of researchers. encodes a novel set of genes that are crucial for antigen presentation, namely the TAP peptide transporters, proteasome subunits, and HLA-DM. Thus, for MHC class I presentation a major focus will be to understand the specificity and mechanism of the TAP transporters and their associated components. This work will have implications far beyond immunology as the transport mechanism of the large family of ABC transporters (including the multi-drug resistance transporter) is totally unknown. For MHC class II presentation the role of the newly discovered HLA-DM molecule will be a major goal as well as the structure of the MHC class II/Invariant chain (Ii) complex from which Ii has to be removed prior to peptide loading, and, importantly, the intracellular transport of class II molecules and identification of endosomal sub compartments where peptide loading takes place. With these efforts it will be possible to provide a fairly complete picture of the intracellular events important for antigen presentation. The results obtained will be important to devise strategies for vaccination and intervention in cancer, auto immune disorders and allergy. The network partners have all participated the work leading up to this project, a project which is based on already preexisting, fruitful collaborations between various network partners.
