Research objectives and content Defects in initiation and execution of apoptosis are common in cancer. They are critical for neoplasticdevelopment and resistance to cytotoxic chemotherapy. This project investigates the reversibility of l this cancer phenotype. The host laboratory has generated tumour suseptible mice by targeted deletionof genes involved in apoptosis. Cell lines have been derived from tumours arising in these mice and partially characterised. By restoring absent components of the cell death pathway, or genes involved downstream, the effect on regulating apoptosis and response to therapy will be studied. Cell lines will be transfected with conditional expression vectors where the transfected gene is expressed in the presence of tamoxifen. The effects on apoptosis in vitro will be studied by direct examination of cells using time lapse videomicroscopy. By injecting cells into mice, the in vivo effects of conditional expression of death genes will be studied using morphometric analyses and flow cytometry. Training content (objective. benefit and expected impact) The training objectives are to gain further experience of molecular biology techniques, particularly the use of novel systems for the conditional expression of genes, and methodology used to assess apoptosis in vitro and in vivo in a major European laboratory working on apoptosis.
