Цел на проекта
The growing knowledge of D. melanogaster genome allows us to answer unsolved questions about genome evolution. One unsolved question is wether the gene arrangement has any biological meaning or merely reflects the history of random inversion fixation. We propose a comparative study of physical and functional evolution of the genome of Drosophila at two different levels: one at the cytogenetic and the other at the molecular level. We propose to study a region that is by a long way the best genetically analysed gene region in Drosophila We want to carry out a characterisation of Adh-Adhr bicistronic genes and any other conserved segment in the repleta group. OBJECTIVES I.-To localise by in situ hybridisation on the polytene chromosome of D. repleta and D. buzzatii all the genes described in the 2 Mb region around the Adh locus (chromosomal sections 34D-36A) of D. melanogaster to estimate the average size of the unbroken fragment and the evolutionary rate of fixation of inversions for Muller's element B. 2.- To characterise at the molecular level the fragment we expect to be conserved: AdhAdhr (bicistronic genes) and any other conserved fragment to study its physical and functional organisation in the repleta species group to asses if the same organisation is conserved in different species of the genus. For D. melanogaster, there exists a huge of information of the functional organisation of this region (34D-36A) and it is important to asses if the same organisation is conserved in different species of the genus. This protocol will allow the more extensive comparative analysis of physical location and function of a region carried out in Drosophlla. Otherwise, the proposed study will provide to Esther Betran a training in Molecular Biology and in Computer Sequence Analysis. She also will have the opportunity to join a group working in one of the biggest projects in Genetics: the Drosophila Genome Project
