Цел на проекта

Research objectives and content A number of species of Leishmania cause a disease spectrum ranging in severity from asymptomatic or mild infections to localised, diffuse or muco-cutaneous disease and visceral leishmaniasis. The final tropism of the parasite and the clinical outcome are most likely to be determined by an as yet unravelled complex interplay of molecules derived from the sandfly transmitting vector, the host immune arsenal molecules and the leishmania parasite itself. The main goal of the proposal is to investigate the gene encoding for an specific virulence factor in Leishmania chagasi or L infantum parasites causing either cutaneous or visceral leishmaniasis. In addition, transcripts would be compared between L chagasi and L infantum isolated from patients with cutaneous lesions or suffering visceral disease. The eventual aim is to gain a better understanding of the nature and role of parasite virulence genetic markers for predicting tropism and clinical outcome. Specific objectives are: 1. To define the organisation, copy number, chromosomal location and expression of the gene LPG1 encoding a galactofuranosyl transferase like enzyme in L. chagasi and L. infantum reference parasites from patients with either VL or CL. 2. To identify unique transcripts or quantitative differences in the relative abundance of transcripts by differential analysis of mRNAs of L. chagasi and L. infantum reference parasites from patients with either V1 or CL. 3. To investigate the role and essentiality of the Leishmania gene identified in 1 or 2, which best correlates with pathology, by functional complementation following Leishmania transfection. Training content (objective; benefit and expected impact) The main training goal is to transfer to the applicant the most complete overall package of state of the art molecular parasitology techniques together with ensuring that he/she obtains a reliable criteria for experimental design, experience in communicating and discussing scientific results (poster and oral presentations, paper writing in reputable journals), grant proposal writing and develops an effective team and network approach in research.
