Цел на проекта
Research objectives and content Recent studies have led to the identification of a new set of proteins (called coactivators) that interact with the nuclear receptors in a ligand-dependent manner playing essential roles in mediating their transcriptional activities. Two of these proteins are SRC-1 (and related proteins) and p300/CBP family. Despite the growing information about these coactivators little is known about how they influence the transcriptional activity. We aim to elucidate the role of SRC-1 and p300/CBP protein families in the transcriptional regulation carried out by nuclear receptors. To do this we propose to: a) Produce SRC-1 and p300/CBP mutants and assay in vivo their abilities to modulate nuclear receptor activity, b) Introduce in intact cells the peptides responsible for the protein interactions testing how they affect the nuclear receptor transcriptional activity. Training content (objective, benefit and expected impact) The completion of this proposal would help to define how ultimately nuclear receptors activate transcription and how these activities are regulated by their ligands, offering new insights into molecular controls of many process in which are involved the nuclear receptor superfamily. In addition many of these receptors and their ligands are associated with known human diseases and our results could have a potential impact in posterior trials looking for therapies for these diseases.
