Research objectives and content Ets2 is a transcription factor regulating the expression of genes involved in cell growth, differentiation and developmental processes. Unpublished results from Dr. Boulukos' laboratory show that Ets2 can also inhibit programmed cell death (apoptosis). This is the first observation indicating that Ets2 can play a role in controlling cell death processes. They observe that while macrophages die by apoptosis in the absence of macrophage colony stimulating factor (CSF-1), a factor necessary for the growth and survival of myeloid cells, macrophages constitutively expressing Ets2 survive under the same conditions. I propose to investigate whether this inhibition of programmed cell death by Ets2 is a general phenomenon. Two different cellular systems available in the laboratory will be used to address this question. I will develop a third cellular system where Ets2 expression will be inducible. This system will be used to determine at what point(s) Ets2 expression is necessary to inhibit the initiation of the cell death program. Finally, I will investigate the mechanism of inhibition of apoptosis by Ets2. Several genes are implicated in controlling cell death. I am interested in determining whether one or several of these genes are direct downstream targets of Ets2. Training content (objective, benefit and expected impact) I chose Dr. Boulukos' laboratory because I want to obtain in experience in a new and exciting field of research. My objectives are to learn about a transcription factor which not only induces macrophage differentiation, but also controls the cell death program. I will clearly benefit by learning the current techniques used in molecular and cell biology including cloning, sequencing, Northern and Western analyses, cell transfections, gel retardation assays, transcription activation assays and immunofluorescence studies. Dr. Boulukos' laboratory has both the equipment and experience to provide an excellent scientific environment for my training as a post doctoral fellow. The impact of this work is of great importance especially considering that deregulating Ets2 expression can result in certain types of leukemias. Links with industry / industrial relevance (22)
