Research objectives and content Members of the fibroblast growth factor (FGF) family are implicated as important cell to cell signalling molecule in animal development. The role of FGF signalling in human development has been further highlighted by the demonstration that mutations in FGF receptor-3 are the major cause of human dwarfism, and mutations in other FGF receptors cause several inherited skeletal dysplasias of children. As targeted deletions of FGF receptor-2 are lethal in the mouse at implantation, we plan to use a more delicate approach where only one of the two alternative FGFR-2 isoforms are mutated in the mouse germline. In addition, the approach will generate other lines of mice where the FGF receptor mutations are restricted to specific cell lineages. This will allow us to investigate the significance of FGF signalling in early mammalian development, organogenesis, and homeostatic mechanisms such as wound repair. Training content (objective, benefit and expected impact) This project will provide training in mouse developmental genetics and mammalian embryogenesis. The results should provide a better understanding of how FGFs affect lineage determination and tissue patterning and may have a direct impact on the understanding of some skeletal dysplasias.
