Цел на проекта
Research objectives and content This proposal concerns the reaction mechanism of the hobo transposon from Drosophila melanogaster. This transposon, and other members of the same family, are unusual because they leave behind a distinctive 'footprint' at the donor site after transposition. The structures of the footprints are not compatible with the gene conversion pathway of double strand break repair which is used to restore the donor site after transposition of other types of elements, such a P in Drosophila and Tcl in Caenorhabditis elegans. The footprints can be explained by postulating a hairpin intermediate on the broken donor ends. However, this mechanism requires the hobo class of transposons to have a reaction mechanism which is significantly different from all other transposons studied to date. I propose to reconstitute the hobo transposition reaction in vitro so that reaction intermediates and products can be examined directly and the reaction mechanism determined. I hope that my research will not only contribute to the basic science of DNA recombination, but that it will also lead to industrial applications. Training content (objective, benefit and expected impact) During the research proposed here, I will receive a thorough education in transposition and DNA recombination in general. I will also learn numerous molecular, biochemical and cell biological methods. The scientific enviromnent and courses offered at Oxford will encourage my development as an independent scientist.
