Цел на проекта
Research objectives and content The primary objective of this research is to bioengineer pancreatic beta cells suitable for mechanistic studies of enzyme regulation and later gene therapy of diabetes. At a first step, molecular biology techniques would be used to correct the impaired glucose-sensing apparatus of the popular RINm5F insulin-producing cell line. This would be achieved by overexpression of the two key elements responsible for beta cell glucose-recognition, glucokinase and the GLUT2 glucose transporter. This surrogate beta cell model, would also prove useful for in-depth studies of the regulation of lactate dehydrogenase activity. The overexpression of lactate dehydrogenase should provide a novel means of elucidating its importance in determining the fate of glucose metabolic flux towards mitochondrial oxidation, which is believed to becritical for insulin stimulus-secretion coupling. This innovative research project should yield a number of important new insights into the pancreatic beta cell stimulus-secretion coupling pathway, in addition to generating novel cell lines useful for gene therapy approaches and available for European experimental diabetes research. Training content (objective, benefit and expected impact) The purpose of this training is to gain practical expertise in a spectrum molecular biology techniques to complement biochemistry methods I acquired during my doctoral research.
