Цел на проекта

Research objectives and content Cell-cell interactions are crucial to patterning and cell differentiation in vertebrate embryos. The Eph-related family of membrane-bound receptor tyrosine kinases and their cognate ligands are among candidates for mediating such cell-cell communication. Expression studies and functional analyses have demonstrated a pivotal role for the Eph signalling during the development and patterning of the central nervous system and spinal cord. The complementarity of receptors and ligands expression patterns in different regions of the embryo together with functional studies, have led to the proposal that Eph signalling acts in establishing boundaries between regions, through mechanisms of repulsion. It is likely that Eph signalling in different morphogenetic processes leads to a common set of cellular responses including control of the cytoskeleton. The aim of the proposed research is to study the functions of the Eph signalling system in mesoderm regionalisation during zebrafish embryogenesis. I aim to analyse whether Eph receptor-ligand mediated repulsion is also used by non-neuronal cells to establish boundaries, such as between the notochord and paraxial mesoderm cells, during primary axis formation. Emphasis will be given in investigating the roles of Eph signalling in mediating cell shape and cell movement changes which occur during convergence and extension movements driving notochord morphogenesis in the zebrafish embryo. Training content (objective, benefit and expected impact) The proposed research will enable me to continue and expand my interest in molecular mechanisms of pattern formation in vertebrates. Most importantly, it gives me the opportunity to acquire knowledge in zebrafish embryology and genetics as well as in new techniques of cell biology. Altogether, this research experience combined with my previous knowledge of mouse genetics, would be extremely beneficial for my future scientific career in Greece.
