Цел на проекта

Research objectives and content Enantiopure epoxides are important intermediates for the pharmaceutical industry. Present production methods rely on kinetic resolution. This causes destruction of the unwanted isomer which leads to unnecessary waste. Better production methods rely on asymmetric synthesis where only one enantiomer is produced in the epoxidation step. Current state of the art is a manganese based catalyst developed by E. Jacobsen (Harvard university). Trials with this catalyst for the industrial production of epoxides led to the conclusion that instability of the catalyst under the epoxidation conditions gave rise to low turnover frequency and hence poor economics of the process. There is therefore a clear need for new stable asymmetric epoxidation catalysts. I propose to prepare catalysts based upon ruthenium complexes made with robust ligands with proven oxidation stability. Two classes of ligands will be explored. The first class is based upon extremely stable triazole dicarboxylic acid, in combination with enantiopure 1,2 diaminocyclohexane. The other ligand class will be based upon enantiopure varieties of 2,2'-bipyridine 3,3'-dicarboxylic acid. With these new catalysts I will explore epoxidation of a range of olefins with industrial relevance, such as substituted styrenes, cinnamic acid derivatives, indenes and chromenes. Training content (objective, benefit and expected impact) Training aspects will be abundantly present in the form of exposure to the industrial context, working in a team that routinely works on similar problems from exploratory research via route scouting and process development till large scale production in the plant. I expect to gain a much better insight in the way in which industrial research works. This will aid me in obtaining employment with the chemical industry Links with industry / industrial relevance (22) DSM, the host institute has a large Fine Chemical Division (Turnover 750 million ECU). Major products are intermediates for the pharmaceutical industry, with a special focus on chiral products. DSM Andeno is a large scale producer of enantiopure epoxides.
