Цел на проекта

Research objectives and content The maize transposable element Ac was previously introduced into the diploid model legume Lotus japonicus and shown to transpose to new positions in the genome. The frequency of germinal excision was found to be similar to the excision frequencies observed in tobacco and maize. A transposon mutagenesis programmedirected at the isolation of symbiotic mutants impaired in development of nitrogen fixing root nodules or root nodule function was therefore initiated. With the genetic evidence for Ac tagging available a mutant arrested at an early developmental stage will be chosen for detailed molecular analysis. In order to study the initiation of the developmental process the mutant arrested at the earliest stage will be chosen. The main objectives of the research project are the following: 1) Phenotypic characterisation of the Nod- mutants available from the Ac mutagenesis programmeand determination of the stage of developmental arrest.2) Detailed genetic characterisation of one Ac tagged L. japonicus Nod-mutant arrested at an early stage of the bacterial-plant interaction initiating nodule development. 3) Cloning and characterisation of the tagged gene, studies of the cellular expression during nodulation. Training content (objective, benefit and expected impact) The objective of the training is the study of plant genes involved on the early nodulation process in the model legume Lotus japunicus. This study will help to achieve a better understanding of the molecular basis of the symbiotic interaction and plant genes controlling nodulation that will provide tools for improving nodulation. Links with industry / industrial relevance (22) The Laboratory of Gene Expression has collaborative projects with industry, part of this involves consultation concerning commercialisation.
