Research objectives and content The main objective is to investigate the pathogenesis and epidemiology of PMWS, and to determine the causal link of porcine circovirus (PCV) with this new syndrome. It includes clinicopathological study of natural cases and experimental reproduction of PMWS by inoculation of PCV, the development of techniques for detection of PCV, and collection of epidemiological information in PMWS-affected farms.Technical milestones are standardisation of an immunocytochemical technique for detection of PCV-antigen, and development of a PCR technique for detection of PCV genome. Sequencing of PMWS-related PCV will allow design ofspecific DNA probes to differentiate PCV derived from PK-15 cells and from PMWS cases. A ELISA technique for PCV antibodies will be standardised, and well as immunocytochemical techniques for porcine monocyte-macrophage markers for its use in formalin-fixed, paraffin-embedded tissues. Training content (objective, benefit and expected impact) The candidate will be Doctorate, and could present at the exam for Diplomat of the ECVP. Trained candidate will be able to work as a pathologist, and to conduct research on animal infectious diseases. Links with industry / industrial relevance (22) The design of vaccines to prevent PCV-infection may be of interest for the vaccine-industry.
