Цел на проекта

Light and growth regulators have major effects on plant morphology thro signal transduction cascades that influence the functioning and architecture of the cytoskeleton. Rhizobium Nod factors also influence the reorganization of the actin cytoskeleton in tips of susceptible root hairs, and this reorganization is induced by activating the inositol phosphate pathway. Using living root hairs as a model system, the link between signal transduction and cytoskeleton driven morphological changes willF be characterized. Actin and actin-polyphosphoinositide (PIP2) binding proteins will be cloned from an available root hair library using heterologous probes. Chimeric constructs with green fluorescent protein (GFP) under the control of a root epidermis specific promoter will be made and introduced into legume roots. The GFP product expressed in eukaryotes is fluorescent and the changes in the actin cytoskeleton and in the location of actin-PIP2 binding proteins induced by Nod factors will be imaged using Confocal Laser Scanning Microscope. Thus, this project will generate knowledge on the mechanism of cytoskeleton driven morphogenesis.
