Цел на проекта
The European strawberry production is of increasing importance, and causes high environment impact due to the high chemical application generally requested. Among the most severe pathogens are species of Collecotrichum and Phytophthora. For both fungi various strains with different degree of virulence and a wide range of susceptibility among varieties exists. Studies on the factors involved in these interactions are useful for the detenction and isolation, through DNA-recombinant techniques, of specific genes involved in defense mechanisms. Using in vivo and in vitro (callus and cell suspension) strawberry varieties, characterised by different susceptibility to the two pathogens, the expression of defence genes induced by specific elicitors (host-cell wall fragments and a toxic lOkD protein purified from P. cactorum culture filtrate), will be analysed. Active proteins will be identified and studied using extracellular extract from resistant cultivar (EER). Genes encoding possible active proteins from EER will be also studied. Recently, the autimicrobial activity conferred by a number of gene constructs has been demonstrated in several species. They are now available for exploitation in fruit crops. Methods for strawberry transformation have been described. Regeneration and genetic transformation protocols in the susceptible strawberry varieties, adapted to the Italian cultivation areas, will be developed and used for the introduction of genes encoding active proteins.
