GSK is a world leader in industrial genetics research, using state of the art equipment housed in a purpose-built £250 million research centre, coupled with an unparalleled consumable reagents budget. GSK's total budget for genetics research for 2001 is over £80 million. The research fellows will receive expert project supervision from leading molecular biologists, drawing from a pool of over 200 UK-based research staff in the fields of genetics, genomics and bioinformatics, and will also have access to GSK's academic collaborators and US-based GSK research scientists. The research training can be divided into three main elements:(i) Population based association studies to identify genes influencing bone density. GSK's in-house research efforts have identified several genes with a potential role in the regulation of bone density. These genes will be screened for single nucleotide polymorphisms, utilizing both public database information and our own internal SNP discovery programme. Any polymorphisms identified will be tested for association with bone density in a cohort of 3000 perimenopausal women.(ii) Functional assessment of polymorphisms in genes influencing bone density . The function of gene variants will be tested directly in cell-based assays, with particular emphasis on polymorphisms in known or putative promoter elements.(iii) Biological and genetic analysis of mouse mutants with bone remodeling deficiencies. In collaboration with the MRC, GSK has generated a large number of novel mouse mutants by chemical mutagenesis; several of these mutants have abnormal bone biology, including osteopenia. Selected mutant lines will be extensively characterized and subjected to linkage mapping and mutant gene identification. Genes responsible for the mutant mouse phenotypes will then be tested for association with bone density in our human cohort.
