Цел на проекта
The atherosclerotic disease is the final outcome of a complex process, which begins with the oxidation follow density lipoproteins (LDL). The oxidised LDL accumulates in the arterial wall triggering a localinflammatory/proliferative process, in which different types of cells take active part: lymphocytes, monocytes/macrophages, smooth muscle cells (SMC), endothelial cells and platelets. Secretary phospholipaseA2 type II (sPLA2) is among the proteins known to modify the Ladles. This enzyme not only is able to do it, butt has been found in high levels associated to the atherosclerotic plaques. High levels of circulating enzyme have also been found in cardiovascular disease patients, as well as in several other inflammatory pathologies. Even though sPLA2 has been considered a risk marker in the coronary disease, the mechanisms that involve this enzyme in inflammatory processes and its relationship with cardiovascular diseases remain unclear. Therefore, the main aim of this project is to unravel the implication of the sPLA2 in the vascular biology and atherosclerotic events. Based on the recent description of an specific sPLA2 receptor in monocots and considering the enzyme as a signalling molecule, we propose the characterization of its involvement in the immune and inflammatory response that take place in arteriosclerosis. We want to study and analyse the signalling pathways regulated by sPLA2, making a special stress on the small Teases, as well as the implication of the receptor-mediated response. For that purpose a knock down system of the receptor will be set-up. Once defined, the study will concentrate on the pharmacological modulation of the pathways triggered bysPLA2.
