Even after decades of research on polychlorinated biphenyls (PCBs), very little is known about their specific metabolism, reactivity of metabolites with proteins, DNA and enzymes involved in biotransformation, gender and species differences and sensitivities and the subsequent induction of cancer. Even less is known about these aspects for polybrominated diphenyl ether (PBDEs), the PCBs of the 1990s and 21st century. Knowledge about these aspects for data based risk assessment; risk prevention and preventive or therapeutic interventions are indispensable. One reason for this lack of data is the unavailability of individual, tagged" congeners that can easily be followed and detected.The main objective is aimed to investigate and determinate metabolic in vivo and in vitro pathways of PCBs and PBDEs. To archive this goal the utilization of fluorine tagged persistent organic pollutants (POPs), i.e. F-PCBs and F-PBDEs as new model and marker compounds will be applied. F-PCBs and F-PBDEs will be applied in specific tests for mutagenicity to show the type and location of damage, direct measurements and chemical identification of the DNA adducts involved. F-PCBs and F-PBDEs and their metabolites may be the very tool that is needed to answer these questions.The project is relevant to the objectives of the work program because it will generate a unique chance to raise the level of research within the ERA in the fields of specific tagging techniques, metabolism, gene induction and bio monitoring of POPs. Specifically, f or the OIF program the achievement of knowledge and experience gained at the outstanding world-class research group of Prof. Dr. L. Robertson of the Department of Occupational and Environmental Health of the University of Iowa is vital. This trend-setting research will enable to establish a career in academia as successful and productive scientist and as well rounded mentor for the future generation of European researchers and will curbing brain drain."
