Цел на проекта

It has become clear that unconventional T cells, play a critical role in cellular immunology. Whilst much work is in progress concerning the molecular characteristics of such cells, and of their human counterparts, we remain conspicuously ignorant of t he cells' specificities.The work will be to apply a system that may identify the ligands of the TCR (T receptor) from the mouse skin and from the mouse and human gut. This work is a natural extension of previous projects in the lab that examined TCR biochemistry, that created critical reagents that the project will utilise (rat basophil leukaemia (RBL) transfected with TCR, recombinant soluble TCRs), further reagent (TCR-sensor system).Three experimental systems using these tools, will permit us to screen epithelial cell line expressing the specific ligand and to subject them to expression cloning. The success of these studies will dramatically improve our picture of tissue-surveillance, and the processes by which normal tissue homeostasis transforms into a situation in which the tissue-associated T cell repertoire becomes activated.The experience profile of Jean-Jacques Mention on IEL background, experiences with confocal microscopy, flow cytometrie can clearly add value to the applied project. He will be trained in advanced techniques in molecular biology like expression of soluble gdTCR.Thus the experience of Jean-Jacques Mention on the one hand and the training possibilities offered at the laboratory of Adrian Hayday on the other hand, are complementary. This will result in a synergistic effect and therefore enhance the scientific excellence of the European Union.The training and experience Jean-Jacques Mention will gain from his participation in the proposed project at the laboratory of Prof. Hayday in London, will certainly contribute to his future career and achievement of professional independence as a researcher.
