Final Activity Report Summary - LEVITATES (Targetting VAP-1 to treat chronic inflammatory liver disease)

The broad objective of the research project has been to facilitate the development of novel therapeutic approaches to treat immune-mediated liver disease. Liver disease of this nature has a number of different origins, such as diabetes and obesity, excessive alcohol consumption and infection, and is a significant socio-economic burden in the developed world which is increasing in prevalence. By studying the mechanisms by which immune cells enter the liver the project researchers have identified a protein called Vascular Adhesion Protein-1 (VAP-1) which appears to be a key player in the inflammatory disease process when excessive numbers of inflammatory cells enter the liver resulting in the build-up of diseased (fibrotic) tissue. A particularly important result of the research project has been to characterise in greater detail the way in which VAP-1 helps cells causing inflammation to enter the liver and how this may contribute to the development of liver fibrosis. This has allowed us to design and test drugs which block VAP-1 function and prevent immune cells entering the liver so reducing the inflammation. When this hypothesis was tested in mouse models of inflammatory liver disease (liver fibrosis) it was shown that blocking VAP-1 with a function blocking antibody (biological drug) reduced the degree of liver damage. The results are expected to lead to the development of novel therapeutic options to treat inflammatory liver disease in man.

Chronic inflammatory disease affects almost one third of the EU population and an increasing proportion of this burden is made up of liver disease. Large increases in death rates from chronic liver disease and cirrhosis have occurred in many EU countries and are predicted to continue. Chronic inflammation and cirrhosis develop in response to liver injury mediated by a wide range of reagents from hepatitis viruses, to alcohol and autoimmune processes.In all cases, persistent inflammation drives the progression and severity of disease but there are currently no effective therapies that modulate or switch off chronic inflammation. There is therefore a pressing need for a greater understanding of the mechanisms of chronic inflammatory liver disease as well as for the development of new therapies. The need is best addressed through action at a Community level to establish and maintain long term collaborative studies aimed, in the long term, at the development of new drugs.The applicants have demonstrated a critical role for the endothelial adhesion molecule VAP-1 in the recruitment of effector leukocytes to the liver and that it is the recruitment of effector leukocytes that sustains chronic liver inflammation. One of the applicants has developed novel therapeutic reagents based on antibodies and small molecule drugs to inhibit VAP-1 function.The current proposal has the objective to consolidate and further develop a partnership between the groups using the expertise in the academic group to explore new avenues in VAP-1 research and test candidate anti-VAP-1 therapies developed by the SME in both human in vitro and animal in vivo studies. This will be facilitated by the exchange of knowledge between the partners. The partnership will also strengthen the European biomedical science base by improving the competitiveness of a European SME.
