Накратко на български
Протеинът алфа-синуклеин се модифицира чрез полусинтетични методи, за да се създадат негови варианти с фосфорилиране. Това помага да се разбере как тези промени влияят върху функцията на протеина при здравето и различните заболявания.

Semisynthesis of caged a-Syn
The primary objective of this project is to develop and optimise semisynthetic approaches that allow site-specific modification of a-synuclein with emphasis on applying these approaches to understand the role of post-translational modifications in regulating a-synuclein function in health and disease. The candidate had already started working on this project before the initiation of this grant and made progress towards completing the Semisynthesis of C-terminal phosphorylated a-synuclein. During the five week period she was supported on this grant, she continued to work on the expression and purification of some of the peptides and recombinant proteins required for preparing thephosphorylated proteins. The last week she spent on organising her materials and preparing a summary report on what she had done.

Цел на проекта
The overall objective of this proposal is to determine the effect of specific disease associated post-translational modifications, more specifically phosphorylation and nitration, on modulating the structural and biochemical properties of a-synuclein. In detail, we plan to assess the effect of these modifications on i) the structure of monomeric and membrane bound a-synuclein; ii) the oliogmerization and fibrillogenesis of a-syn; iii) subcellular localization and targeting of a-syn to specific cellular compartments. Towards this goal, we plan to use a combination of chemical biology and semisynthetic strategies to 1) synthesize a-syn variants that are modified at specific sites 2) make use of caged amino acids and fluorescent tags to allow temporal and spatial induction and control of post-translational modification in living cells.
