Periodic Report Summary 1 - MSOT (Quantitative real-time multi-spectral optoacoustic tomography)

The two major goals of this project are: 1) to design a new optoacoustic system for the intravascular system using multi-spectral-optoacoustic-tomography (MSOT) in the cardiovascular system and 2) improving quantification in image formation by developing appropriate reconstruction algorithms. Ad 1) a fiber light-delivery system for near-infrared-fluorescent (NIRF) imaging was designed and built. The system was used in several imaging studies for cardiovascular inflammatory diseases, specifically atherosclerosis. The results of these experiments so far can be used for assessing the required sensitivity for an acoustic detector to perform intravascular MSOT for imaging inflammatory processes. Several mechanical designs were tested for the fiber catheter resulting in a robust system with smooth rotation and translation characteristics. Ad ii) we developed a new improved algorithm for optoacoustic image reconstruction termed interpolated-model-matrix inversion (IMMI) which already was successfully tested ex vivo in mice and in vivo for imaging zebrafish.

Multi-spectral optoacoustic tomography (MSOT) is a biomedical imaging modality which offers a combination of molecular sensitivity, penetration depth, and resolution which are unparalleled in the field biomedical instrumentation. Nonetheless, the long acquisition time associated with MSOT and difficulties in quantifying the images it produces currently limit its possible applications. The aim of the proposed project is developing quantitative high-resolution real-time MSOT techniques for in-vivo molecular imaging in entire animals and potentially in humans. Specifically, the project will focus on applying MSOT for molecular imaging of the cardiovascular system. The suggested research is inherently multidisciplinary and involves optics, acoustic, signal processing, inverse-scattering theory, biology and medicine. The ultimate goal is to bring MSOT closer to the technological maturity required for biological research and medical diagnosis.
